Abstract
The electrochemical immunosensors based on functional nanoparticle (NP) labels were described. We have developed functionalized silica NPs conjugates and functionalized gold nanoparticles for sensitive immunoassay of proteins such as mouse IgG and tumor necrosis factor-alpha (TNF-alpha). These functionalized NPs were characterized by AFM, TEM, X-ray photoelectron spectroscopy, or electrochemistry. And the analytical performance of these immunosensors was examined. It is found that these methods are very sensitive and the limit of detection was found to be down to 1.0 pM level. The work demonstrates the feasibility of developing a cheap, sensitive, and portable device for multiplexed diagnosis of different proteins. This method is simple, selective and reproducible for trace protein analysis and can be extended to study protein/protein, peptide/protein, and DNA/ protein interactions.
