The Sequel II System Sequencer is a high-throughput DNA sequencer machine developed and manufactured by PacBio, and is designed for high throughput, production-scale sequencing laboratories. Originally released in 2015, the Sequel system provides Single Molecule, Real-Time (SMRT) sequencing core technology powering long-read sequencing platform services for a variety of applications across life science community investigations. For projects seeking rapid and cost-effective de novo genome assembly and structural variation analysis, the PacBio HiFi technology is developed for sequencing up to 8 SMRT Cells in a single run, starting at just 5 ng. HiFi reads are produced using circular consensus sequencing (CCS) mode on PacBio long-read systems, and provide base-level resolution with 99.9% single-molecule read accuracy (Wenger, A. M., et al. 2019). PacBio open-source SMRT Analysis software suite is designed for use with SMRT sequencing data and is provided through PacBio. Primary applications include whole genome sequencing for de novo assembly, metagenome sequencing, RNA targeted analysis, epigenetic characterization, variant detection and many other services for comprehensive views of human, plant, animal, and microbial genomes, transcriptomes, and epigenomes applicable to both biomedical and environmental research topic investigations.
PacBio Sequel II pre-processed raw data is packaged in a .fastq format and can be accessed at project dataset download DOI containing additional processed data products, metadata, and software analysis outputs related to a long read sequence analysis.
